Pemetrexed
Indications :
- Mesothelioma (in combination with cisplatin)
- NSCLC
Contraindications:
Dosing :
- Coadministered with low-dose folate and B12 supplements to prevent toxicity
MoA :
- Blocks folate usage for production of purine and pyrimidine nucleotides, inhibiting DNA replication
- Thymidylate synthetase (TS)
- Dihydrofolate reductase (DHFR)
- Glycinamide ribonucleotide formyltransferase (GARFT)
- Similar to Methotrexate, but blocks more enzymes
